I would like to grow anaerobic bacteria in a liquid medium, but I don´t know exactly how to do it, as I have never worked with bacteria before.
I have an anaerobiosis jar that it can be used once anaerobic bacteria have been inoculated in Petri Dishes, if I´m not wrong the only thing I have to do is to introduce the Petri Dish in the jar and incubate it at the adequate conditions. However, my problem is that I want to inoculate the bacteria not only in agar but also in broth medium, in order to afterwards, take some aliquots from there and freeze the bacteria in 20% of glycerol (I will receive liophylized bacteria and will resuspend them, and I want to make as many aliquots as it´s possible to freeze them in cryovials). So, is it possible to take an inoculate of the resuspended bacteria, introduce it in a liquid medium (I was going to take a 50 ml Falcon tube and fill it with more or less 20 ml of liquid medium) and the introduce this Falcon tube in the anaerobiosis jar for incubation together with Petri Dishes?
I´m conscious I should be fast in the manipulation and that I need to do it in aseptic conditions (I only have a bunsen lighter), as anaerobic bacteria are sensitive to the Oxygen, and I would really appreciate if somebody tell me if what I have asked is possible to do it and if anyone could give me any advice about it.
