Neuro2a differentiation by retinoic acid

Postby nurdiana.anuar » May 02 2012 6:59 pm

Hi

I have started differentiated mouse neuroblastoma Neuro2a (ATCC CCL-131) with 40 microM of retinoic acid for 6 days. I need to assess the differentiation marker in qPCR. Does anyone have any suggestion on the markers that I can use, or any paper that have a list of established qPCR marker for neuro2a RA-induced ? I have been searching for such papers, but most of them use antibody (NeuN, Tuj1) for differentiation assessment. Not many that have a pool of genes for that can be used to assess the differentiation state oin qPCR. I'd appreciate if anyone can give me any advice on this matter. Thank you.

Just use the same target as the antibodies: examples: NEFL, NEFM and NEFH (NM_010910, NM_008691, NM_010904)

Make sure you use the proper negative controls (and if possible positive) to compare with. You should also look up if there is any information available about reliable housekeeping genes in N2A.
