Everybody knows that the next generation sequencing method is a very expensive technique. Actually, I have 7 groups of samples; each group contains about 20 samples.
How many samples are necessary from each group in order to have statistically reliable data? And what's about sample pooling?

Reliable sequence can be obtained when you sequence both strands. DNA sequence cannot lie, and you need not use statistics to prove it is reliable.
If you are looking for the probability of certain mutation in a general population, then you will need statistics.
