Currently trying to write a protocol/research proposal in order to see why a transcription factor activates co-repressors/co-activators, when bound to different promoter regions. It located in the same cell

We hypothesised it may be due to either ligand interaction with the TF or difference in promoter sequencing

We are going to use experiments testing the ligand interaction hypothesis, would we just construct a a library of ligands using a High throughput screening and then check if they are able to bind interact with the promoters and TFs and then do a microarray? Not really sure on what would be the most efficient way to go about doing this
