IRES-sequence, something similar?

Postby Antibes » Jul 27 2012 12:37 pm

Hi, I've got a question and I was hoping someone could help me. For my research proposal (undergraduate student) I'm suggesting a variety of contructs for transfection. I would like to combine my protein of interest with a truncated NGFR, to be able to select my transfected cells with an antibody. So I would just use a construct with an IRES in between, that's fine. However, I have two proteins of interest and I would also like to make a combined construct with the two proteins and the NGFR: proteinA_IRES_proteinB_IRES_NGFR, but I guess I can't use the IRES twice in the same constuct? My superviser said something about a similar sequence for in between my two proteins, something called N2 or N2a. But I can't find anything about it! Does anyone know what he would be talking about?

Thanks!! That is exactly what I was looking for, but it was very difficult to find if you don't know exactly what you're looking for.. You might just have saved my honours certificate.. ;) Thank you so much!!
