STE extraction for Wolbachia detection

When I tested the Wolbachia of Tribolium confusum adults using STE, I got nothing. But it can works for pupae.When I tested the other beetle(Larsioderma serricorne) using STE, it can works for adults and pupae. Using columns for extraction, pupae and adults of both beetle species test positive. I have obtained this result several times.I will appraciate if anyone can tell me the probable reason or maybe someone have the same problem with my. I am look foward to get the answer.
